Sanger sequencing, fragment analysis


The institute acquired a 454/Roche GS FLX sequencing instrument through funding from the Prefecture of Crete. This state-of-the art instrument now forms part of IMBBC's well equipped genome sequencing platform. 

The hallmarks of 454 Sequencing™ are its simple, unbiased sample preparation and long, highly accurate sequence reads, including paired reads. 454's current system, the Genome Sequencer FLX with GS FLX Titanium series reagents generates 400 million high quality bases per 10 hour instrument run. GS-FLX 454 Sequencing technology has enabled hundreds of peer-reviewed studies in various research fields such as cancer and infectious disease research, drug discovery, marine biology, anthropology, paleontology to name but a few.

Recently the institute acquired a GS Junior sequencing instrument. The instrument shares the characteristics of the larger GS-FLX, but with an output and cost of 1/10th, and simpler sample preparation and shorter run times.

  1. Whole genomes (Plant, animal, bacterial etc)
  2. BACs, YACs, Fosmids, viruses etc
  3. Transcriptome (cDNA, ESTs)
  4. Metagenomes
  5. Large PCR products (>1.5 kbp)
  • Amplicon sequencing of PCR fragments

  • Paired End sequencing

Sanger sequencing, fragment analysis 

The institute acquired an ABI 3730 automated sequencing instrument through funding from the MARBIGEN project. The sequencer is equipped with 48 capillaries of 36 cm length. 


The ABI 3730 sequencer is regarded as the "Gold Standard" in Sanger sequencing. The instrument can be used for Sanger sequencing, or fragment analysis (e.g. microsatellites, AFLP, SNP genotyping). Applications include e.g. species identification, SNP screening, mutation detection, parentage analysis, population genetic and phylogenetic studies.
